Skype unfortunately does not use a single port. It uses almost any port 
that it can to get past NAT and firewall configurations. In a similar 
way to other peer-to-peer applications like bittorrent.

You would need to do layer 7 filtering with a suitably capable firewall. 
You also need to do layer 7 inspection on port 80 & 443 traffic as Skype 
is even capable of proxying via HTTP.
